PRECISION DIAGNOSTICS

Purpose/Principle

To rule out the presence of maternal cell contamination within a fetal sample.

Special Collection Notes

Collect specimen according to standard operating procedure.

Specimen Type

-Whole Blood/Bone Marrow :

Purple Top Tube-EDTA

 

-Amniotic Fluid/CVS (Cell Cultured):

Two T25 Flask, filled with media

 

-Amniotic Fluid/CVS(Direct):

Sterile screw capped container

*Please establish a cell culture before sending direct fluid

 

-Tissue (Fresh Frozen, Paraffin Embedded, or RPMI)

Specimen Volume

Whole Blood/Bone Marrow

Preferred: 4.0 mL

Minimum: 0.5 mL

*Receipt of blood or bone marrow within 72 hours of collection. Delays may impact nucleic acid quality.

 

Tissue

Preferred: uncut paraffin-embedded tissue block. Slides: 1 stained and 10 unstained - 10 micron thick slides

*Receipt within 24 hours of cutting or as soon as possible. Delays may impact nucleic acid quality.

 

Amniotic Fluid (Direct):

-For second trimester samples, send two (2) samples of direct amniotic fluid, 15–20 ml each, in sterile, securely capped plastic tubes.

-For third trimester samples, send a total of 45–50 ml of direct fluid. Transport promptly at room temperature.

 

Amniotic Fluid/CVS(Cell Culture):

Two T25 flasks, filled with media for shipment.

 

CVS (Direct):

At least 10–15 mg of tissue in a sterile medium in a sterile, securely capped plastic container or CENTRIFUGE tube.

Processing Instructions

Store and ship ambient. If transport is longer than 4 hours send refrigerated on cold pack. For frozen tissue please send on dry ice- ASAP. CVS and Living Cells in Tissue Culture Flask or Amniotic Fluid - Must be store at Room Temperature.

 

NOTE: Samples should be shipped after collection as soon as possible. Ship samples for arrival at Children’s Hospital Colorado on Monday through Thursday. Our recommendation is to use FedEx or UPS for shipping and obtain a tracking number to ensure safe delivery. Samples that are received on a Friday afternoon, Saturday, Sunday or before a major holiday will not be processed until the next business day. If samples need to be stored in your laboratory prior to shipping, they should be stored at room temperature for no more than 72 hours.
