Mitochondrial Lab

Purpose/Principle

Pyruvate dehydrogenase complex (PDHC) is located in the mitochondrial matrix and catalyzes the oxidative decarboxylation of pyruvate. This results in the formation of acetyl CoA and CO2. Acetyl CoA is then oxidized in the Krebs cycle.

Special Collection Notes

Skin Punch:
4 mm skin punch collected using aseptic technique and transported in a sterile container such as a 15 mL conical tube filled with transport media or tissue culture media.

T-25 Flasks x2

Processing Instructions

Skin Punch: Ship overnight in a Styrofoam box on wet ice

 

Fibroblast: Send two T-25 flasks containing confluent fibroblasts (with plug-seal caps, completely filled with media) in a Styrofoam box at room temperature. Fibroblasts must be mycoplasma free and grown only in fetal calf serum of US origin (please provide documentation of any previous mycoplasma testing results). Include the passage number (if known) and any relevant information about the patient.
