Test Code LAB7078 Blue Native Electrophoresis, Fibroblast

Performing Section

Mitochondrial Lab

Purpose/Principle

Using the current method, four different complexes are assayed in fibroblasts: complex I, II, IV, and V . After isolation of the mitochondrial membranes containing the electron transport complexes, the complexes are separated on a non-denaturing blue native PAGE gel. This separates the complexes from each other and from other interfering enzyme activities that typically complicate the interpretation of classic enzyme activity analysis in whole homogenates. The gel is then cut into four or five parts if assaying complex III activity, and the activity of each complex is analyzed by an in-gel enzyme activity with a color end product. The activity is analyzed by comparison to the same assay of control tissue run on the same gel prepared in the same batch. The assay is qualitative.

Special Collection Notes

Skin Punch:
4 mm skin punch collected using aseptic technique and transported in a sterile container such as a 15 mL conical tube filled with transport media or tissue culture media.

Minimum: 4mm

Processing Instructions

Skin Punch: Ship overnight in a Styrofoam box on wet ice

Fibroblast: Send two T-25 flasks containing confluent fibroblasts (with plug-seal caps, completely filled with media) in a Styrofoam box at room temperature. Fibroblasts must be mycoplasma free and grown only in fetal calf serum of US origin (please provide documentation of any previous mycoplasma testing results). Include the passage number (if known) and any relevant information about the patient.
